Ligation reactions were set up as follows:
-Reaction x 3+ve control
Digestion product (4712/63)4 μL0 μL
pBAD/His4 μL4 μL
Water0 μL4 μL
Ligase buffer1 μL1 μL
Ligase1 μL1 μL


The reactions were placed in the cold room at 4 °C overnight.
Digestion product and pBad/His are at reasonably even concentrations.

I have omitted the +ve control as what I was intending to do would be nonsense: the pBAD/His would not ligate to itself as it was cut at two sites not one. This reaction in itself is technically the positive control as the whole cycle is being done in aid of checking as a positive control.
